General application

De‐Plasma™ is designed to prevent Mycoplasma contamination in all types of cultured cells. It can also be used to decontaminate cell culture that has already been infected with Mycoplasma.


De‐Plasma™ is an antibiotic cocktail comprised of one macrolide, one quinolone and one tetracycline compound in a single vial. This three compound mixture offers lower cytotoxicity and higher potency compared to other products on the market. It is effective against all known Mycoplasma species and related non‐cellwalled prokaryotes.

Mode of Action

The mode of action for each of the antibiotics in De‐ Plasma™ is different. Tetracyclines and macrolides block protein synthesis by interfering with translation at the prokaryotic ribosome. Quinolones inhibit bacterial DNA gyrase and topoisomerase IV, blocking DNA replication. Eukaryotic cells do not contain DNA gyrase or topoisomerase IV and are therefore unaffected by De‐ Plasma™ treatment.


De‐Plasma™ can be added directly to the bottle of culture medium or to the flask containing the cells. It is recommended that the entire contents of one 2.5 mg vial of De‐Plasma™ be added to 500 ml of fresh media. De‐Plasma™ can be stored in solution at 4°C for up to 3 months.

Maintenance or prophylactic concentration of De‐Plasma™ A: 5 μg/ml.


  • Split an actively dividing culture of cells into media containing 5 μg/ml of De‐Plasma™ A. (2.5 mg De‐Plasma™ A per 500 ml of media)
  • For maintenance of Mycoplasma free cultures, continue using De‐Plasma™ A at a concentration of 5 μg/ml
Cultures can be checked for Mycoplasma contamination using TOKU‐E’s PCR Mycoplasma Detection Kit.
